                  Visualization of Viscous Fingering Phenomena During Chromatofocusing


    Illustration of fingering phenomena in a concentrated band of protein focused on a retained pH transition. Experiment was performed using a 26 x 1 cm glass column packing with 35-µm Sepharose HP medium pressure stationary phase obtained from Amersham Pharmacia Biotech. The column was presaturated with an acetate buffer at pH 5.5 and eluted with an acetate-formate buffer at pH 4.5. The band contained approximately 20 mg of GFP. Buffers were prepared with 0.05 M NaOH and the flow rate was 2.0 ml/min. The upper photograph was taken soon after feed introduction while the lower photograph was taken when the GFP band had migrated approximately 50% of the column length and indicates the presence of fingering.  Flow direction is downward.
